The group is a criminal organization based off astrology and are longtime villains of SHIELD. Each member has his/her base in a different American city. As the stars change a different sign appears in the sky the corresponding Zodiak member takes control of the group. They want to control and dominate the political and economic landscape of the United States and will use any means to accomplish their ends.

Each member has identity hidden by a mask and a costume. Each costume is in the theme of the constellation that he/she represents. There have been many different iterations of the cabal with different signs running the show.

Scorpio:
The Zodiac member Scorpio stands out in importance. In the comics Scorpio has his run ins with Nick Fury and later turns out to be Jake Fury, Nick Fury's diabolical brother. (This is later retconned so that he is a double agent). He is also able to infiltrate SHIELD pretending to be his brother.
The group has an artifact called the Zodiac Key which gives them massive multidimensional powers.
Marvel Cinematic Universe:
The Zodiac is mentioned in the Marvel One Shot " Agent Carter. Agent Carter single handedly retrieves a serum controlled by the Zodiac Group.
It is not known whether the group will return in the Agent Carter series or in modern time based Agents of SHIELD show.
